I'm going to correct Claudio on one thing. The statue is Tawaret, not Sobek (though sometimes she is depicted with Sobek on her back). This is confirmed in the abc.com episode recap.
I arrived at the same conclusion as Waldorf and Sauron: Jacob's nemesis (Esau?) is the Black Smoke Monster. Furthermore, he was being held captive in the cabin, not Jacob. That's why we see Christian Shepherd there, several times. And he was finally able to escape when the circle of ash was broken. Also, in the first scene, we see Jacob and his nemesis argue about whether to let The Black Rock land on the island. The nemesis doesn't want them to land—he wants to protect the island, just like the Black Smoke Monster is a security system for the island.
